Description
Semax (Met-Glu-His-Phe-Pro-Gly-Pro) is a heptapeptide analogue of the ACTH(4-7) fragment that lacks the hormonal activity of full ACTH but retains the neurotrophin-stimulating effects. Its primary mechanism is upregulation of BDNF (brain-derived neurotrophic factor) and NGF (nerve growth factor) in the hippocampus and frontal cortex — the regions most involved in working memory, attention, and executive function. It also modulates dopaminergic and serotonergic transmission and has demonstrated neuroprotective effects in ischemia and traumatic brain injury models.
For women, Semax is relevant because the cognitive symptoms most reported in perimenopause — word-finding difficulty, working memory lapses, slowed processing speed, and the generalized “brain fog” that can be disabling — are mechanistically linked to declining BDNF signalling (estrogen normally supports BDNF expression in the hippocampus). Semax provides a direct BDNF-stimulating signal that operates independently of estrogen, offering a viable approach to cognitive support during hormonal transition. Research shows improved performance on attention, learning, and memory tasks, with onset within 15–30 minutes of intranasal administration.
