Description

MOTS-c (mitochondrial open reading frame of the 12S rRNA type-c) is a 16-amino acid peptide encoded in the 12S rRNA gene of the mitochondrial genome — one of a class of peptides called “mitokines” that allow mitochondria to communicate with the nucleus, cytoplasm, and distant tissues. Its primary mechanism is AMPK activation — the cellular energy sensor that responds to energetic stress by increasing glucose uptake, promoting fat oxidation, and inhibiting anabolic processes that consume energy. This is precisely the metabolic shift that aerobic exercise produces.

In female biology, MOTS-c is particularly relevant because AMPK activity declines with age and is suppressed by high estrogen — meaning women naturally have lower MOTS-c/AMPK activity in reproductive years, with metabolic consequences that become apparent after menopause. Research in animal models shows MOTS-c administration reverses diet-induced insulin resistance, reduces fat accumulation, and extends healthspan. In human cells, it has demonstrated protection against metabolic stress and inflammation. It is also being studied for its role in protecting oocyte quality — an emerging area of female longevity medicine.

Female-calibrated dosing: 5–10mg subcutaneously 3x/week. Women typically respond at 5mg. Best administered in the morning or before exercise to leverage its exercise-mimetic effects. Cycle 8–12 weeks.
